La diferencia está en la concentración de yodo.

• El Lugol al 2% contiene 2% de yodo y 4% de yoduro de potasio. O lo que es lo mismo ~2 g (≈ 20 mg/mL) de yodo elemental y ~4 g de yoduro de potasio por cada 100 mL de solución.
  
• El Lugol al 5% contiene 5% de yodo y 10% de yoduro de potasio, es más concentrado y potente. O lo que es lo mismo: ~5 g de yodo elemental (≈ 50 mg/mL) y ~10 g de yoduro de potasio por cada 100 mL.
  

En resumen: el 5% tiene más del doble de yodo que el 2%, por lo que ofrece una acción más fuerte.
