The difference lies in the concentration of iodine.

• 2% Lugol’s solution contains 2% iodine and 4% potassium iodide. In other words, ~2 g (≈ 20 mg/mL) of elemental iodine and ~4 g of potassium iodide per 100 mL of solution.
• 5% Lugol’s solution contains 5% iodine and 10% potassium iodide; it is more concentrated and potent. In other words: ~5 g of elemental iodine (≈ 50 mg/mL) and ~10 g of potassium iodide per 100 mL.

In summary: the 5% solution contains more than twice as much iodine as the 2% solution, and therefore has a stronger effect.
